Born in Ogun State, Bankole was the fourth of six children born to architect Babatunde Bankole and administrative secretary Titilayo Bankole. Taiwo, her twin sister, also acts from time to time. She also starred alongside Toyin Abraham in the film Prophetess.

Additionally, she received the prestigious Africa Magic Viewers’ Choice Award (AMVCA) for best actress. She won the 2015 Best Actress in Drama award for her performance in October 1 by Kunle Afolayan. Bankole’s role in the popular film Adire earned her the Best Lead Actress prize in the 2024 edition.
